THE USE OF INTERFERON (IFN) IN THE TREATMENT OF ACQUIRED IMMUNE-DEFICIENCY SYNDROME (AIDS) RELATED KAPOSI'S SARCOMA (K.S.). ADAN

We report here the results of three sequential clinical trials in 49 patients with AIDS-related K.S. initiated in March 1982. Leukocyte IFN (IFNα) were used at a dose of 3 x 106 Mµ i.m. daily x 30 days; Lymphoblastoid IFN HuIFNα (Ly) 20 x 106 Mµ/m2 i.e daily x 60 days alone and in combination with vinblastine 5 mg/m2 IV q.o. week. Response rates were:

The median duration of responses has been 28+ weeks (range 7-36 weeks) for HuIFNα (Ly) and 9+ weeks (range 8-24+ weeks) for HuIFNα (Ly) + vinblastine. Clinical toxicity and myelosuppression were marked in the patients receiving the combination treatment. We conclude that low dose IFNα has no antitumor activity. HuIFNα (Ly) at doses > 20 Mµ/m2 has activity in patients with AIDS related K.S. alone and in combination with vinblastine. Further studies are indicated to improve the therapy of this disease.
