A radioimmunoprecipitation assay (RIP) to a core protein (p25) of LAV, a Western blot (WB) against p41 and/or p24 of HTLV-III after an ELISA screening and an ELISA against HTLV-III/LAV CDC451 virus were each used to test for retroviral serum antibiodies in 25 couples consisting of index Pts with AIDS (21) or AIDS-related complex (ARC) with oral candidiasis (4) and their HP with no other risk factors. Partners had interviews, physical exams and lab testing performed every 4 months in a prospective study (mean follow-up 19 mos. from the first symptom in the index patient). Controls included 85 NYC blood donors, 184 hospital employees, and 50 lupus erythematosis (SLE) Pts. HP were seropositive more often than controls (P<0.001). One HP developed AIDS and 3 ARC.

T4/T8 ratios in LAV-RIP or HTLV-III (WB) seropositive HP were significantly lower than in seronegative HP (P<0.001). This study shows that antibodies to HTLV-III/LAV occur often in HP of Pts with AIDS or ARC and is consistent with a causal role for HTLV-III/LAV in AIDS and transmission between HP.
