Comparison of various drugs effect on HIV reproduction on primary and chronically infected cell lines.

OBJECTIVE: Two stage scheme of anti-HIV drugs screening is proposed. The first stage is performed on primary infected cells (line MT-4 or peripheral blood mononuclear cells). For the second stage assay a cell line chronically infected by HIV-1 and contained provirus is used.

METHODS: The level of HIV reproduction was examined by measuring of reverse transcriptase activity. Virus antigen expression and percentage of infected cells were determined by immunoenzyme and immunofluorescent methods, respectively.

RESULTS: Comparison of drugs with different mechanisms of action on HIV reproduction was performed. Simultaneously the viability of treated cells was determined. On primary HIV-1 infected cells the virus inhibiting effects were for azidothymidine - 90-97%, heparin - 98-99%, glycyrrhizic acid - 95-97%, antisense oligonucleotides - 80-90%, synthetic peptides from binding site of gp120 - 60-70%. On chronically infected cell line the curing effect of these drugs decreased in different extent. Combinations of drugs with various mechanisms of action were also examined.

CONCLUSION: The drugs and their combinations with clear inhibitory effect on both stages of the screening are perspectively for AIDS treatment.
