Characterization of HIV-1 dual infection in patients of Rio de Janeiro.

ISSUE: To analyse the HIV-1 protease and p24 gag genes in potentially dual infected HIV-positive patients.

METHODS: Viral protease and p24gag genes were amplified by nested PCR, The amplified products of protease were digested with AluI,HinfI, BclI and ScaI. p24gag products were didested with DraI, AluI and HaeIII.

RESULTS: The analyse of such patients showed a dual restriction pattern whendigested with AluI. Then, they, were digested with HinfI, BclI and ScaI showing the expected dual pattern. The same results were obtained when p24gag fragments analysed with DraI, HaeIII and AluI. The PCR products of the protease and p24gag were cloned and sequenced. It was observed that in one patient the recombination between two subtypes of HIV occurred and interestingly we could recover the polar virus which were the source of the proposed viral recombination.

CONCLUSIONS: Use of restriction endonucleases allowed us to identify patients bearing two distinct sequences in the protease gene. The restriction pattern of protease gene suggested that they were infected with subtypes B and C, in three cases, and C and D in another case.
