OBJECTIVE: To study and evaluate the combined screening method (ELISA and particle agglutination) for HIV infection comparing with confirmatory method (Western blot)Method: 173 routinely tested HIV seropositive samples were retrospectively studied by sampling from the sample obtained during April 1993 to December 1994 at Immunology section, Bamrasnaradura Infectious Disease Hospital. These samples were HIV seropositive tested by combined screening method: ELISA (Genelavia Mixt, Sanofi Pasteur) and PA (Serodia - HIV). The confirmatory method: Western blot (Diagnostic Pasteur) was the additional test done in order to compare the results obtained from these two methods.
RESULTS: Of 173 HIV seropositive samples, 160 were both ELISA and PA positive, the rest of 13 were either ELISA or PA positive. From 160 positive samples, 154 (96.25%) showed strongly positive by WB, while only 6 (3.75%) were indeterminate results. Using optical density index (OI) of ELISA calculated by OD. Sample/OD. cut off, all 154 strongly positive by WB showed OI over 10 and also did the 4 indeterminate results. Of 13 either ELISA or PA positive, all had OI of ELISA less than 10. None of them showed positively by WB, 6 of them showed doubtful results.
CONCLUSION: These data showed that the samples tested positively by PA and ELISA test with OI over 10 were not necessary confirmed by WB which was so expensive and seemed to be less sensitive to ELISA and PA.
