OBJECTIVE: To determine and compare the prophylactic efficacies of intermittent doses of ATQ and TMP-SMZ for Pneumocystis carinii pneumonitis.
METHODS: Sprague-Dawley rats were immunosuppressed with dexamethasone x 6 weeks to provoke PCP. Groups of 10 or 11 rats received no drug (control) ATQ 100 mg/kg or TMP-SMZ 50/250 mg/kg orally daily, 3 days per week (MTW), weekly or biweekly or no drugs (control). At 6 weeks necropsied lungs were studied for PCP.
RESULTS: Of the 91 animals studied 86 were evaluable at the completion of the experiment. Results are tabulated: (table: see text)
CONCLUSIONS: Both ATQ and TMP-SMZ prevent PCP in 100% of animals when given daily or 3 days per week and in 80% when given once weekly. One dose of ATQ every 2 weeks was ineffective while TMP-SMZ every 2 weeks prevented PCP in 60% compared to only 9% of controls with no PCP.
