BACKGROUND: To assess and characterise changes in expression of IL-4 and IFN-g on CD4+ T lymphocytes during HIV-1 infection in patients without therapy, and after initiation of HAART.
METHODS: We followed 13 HIV-positive patients, who were treated at least 6 months by HAART and 4 HIV-positive patients without therapy. In the control group we selected 7 HIV-negative healthy persons.Flow cytometric analysis was employed for direct detection of intracellular cytokines visualized by fluorochrome labeled monoclonal antibodies.
RESULTS: In HIV patients without therapy minimal expression of both examined cytokines was noted (IL-4: 0.25% without, 0.42% after stimulation, IFN-g: 0.19% without, 0.59% after stimulation), compared to normal healthy volunteers (IL-4: 0.63% without, 3, 75% after stimulation, IFN-g: 0,46% without, 13,37% after stimulation). Group of HAART- treated patients, where treatment lasted at least 6 months, showed significant elevation of both cytokines expression (IL-4: 1,19% without, 4,08% after stimulation, IFN-g: 0,72%without, 5,65% after stimulation).
CONCLUSION: Our results demonstrate significant elevated expression of Th-1 (IFN-g) and Th-2 (IL-4) cytokines in a group of HIV patients treated by HAART at least 6 months, when compare to the HIV patients without therapy. IL-4 production was slightly higher in patients with HAART than in HIV negative control group.Long-lasting expression of cytokines during HAART is probably welcome sign of the immune system reconstitution.
