Antibody-mediated neutralization against dual tropic viruses depends on coreceptor usages.

BACKGROUND: It is said that neutralization against human immunodeficiency virus type 1 (HIV-1) is unaffected by the coreceptor used in previous reports. Now we report the evidence that antibody-mediated neutralization against dual tropic viruses depends on coreceptor usages when we used monoclonal antibodies and coreceptor-restricted cell lines as neutralizing target cells.

METHODS: We used U373-MAGI series and GHOST series as target cells of neutralization assay. Three dual tropic, one X4, two R5 strains and envelope-pseudotyped viruses were tested. Eight monoclonal antibodies against HIV-1 envelope glycoprotein were used. The binding activity of monoclonal antibodies against HIV-1 was determined using flow cytometric analysis.

RESULTS: Two monoclonal antibodies, K2-28 and r21, neutralized the infection of dual tropic viruses, KMT1 (primary isolate) and 89.6, to CCR5 expressed target cells, but not to CXCR4 expressed ones. Other six antibodies did not show such differences. r21 recognizes V3 region of env. K2-28 was developed using recombinant gp120 from SF162 and showed different patterns between neutralization and binding activities against R5 tropic HIV-1.

CONCLUSIONS: We showed the evidence that antibody-mediated neutralization depends on coreceptor usages, and the different role of antibodies between neutralization and binding against HIV-1.
